[摘要]
【目的】观察电针足三里、天枢、三阴交、中脘穴对高脂饮食诱导的肥胖小鼠的治疗作用及机制。【方法】通过饲喂高 脂饲料建立肥胖小鼠模型,将其随机分成模型组,电针组(针刺足三里、天枢、三阴交、中脘穴),电针+抑制剂组(针刺足 三里、天枢、三阴交、中脘穴,腹腔注射LY294002),另将饲喂普通饲料的小鼠作为正常组。干预结束后,测定各组小鼠体 质量,全自动生化分析仪测定空腹血糖、总胆固醇、甘油三酯、低密度脂蛋白胆固醇,苏木精-伊红(HE)染色法观察肝脏组 织形态变化,酶联免疫吸附分析(ELISA)检测肝脏组织促炎因子白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)、白细胞介素 1β(IL-1β)及脂质过氧化产物丙二醛(MDA)水平,比色法检测肝脏组织抗氧化酶超氧化物歧化酶(SOD)、过氧化氢酶(CAT), Western Blot 法检测肝脏组织中磷脂酰肌醇3-激酶(PI3K)、磷酸化(p)-PI3K、蛋白激酶B(Akt)、磷酸化(p)-Akt、p65、磷酸 化(p)-p65 蛋白表达水平。【结果】与正常组比较,模型组小鼠肝组织结构破坏严重,肝细胞内有空泡,并出现肿胀,体质量,空腹血糖、总胆固醇、甘油三酯、低密度脂蛋白胆固醇,IL-6、TNF-α、IL-1β、MDA 水平和p-p65 蛋白表达量增加, SOD、CAT 活性及p-PI3K、p-Akt 蛋白表达量降低,差异均有统计学意义(P<0.05);与模型组比较,电针组小鼠肝脏组织 结构改善明显,体质量,空腹血糖、总胆固醇、甘油三酯和低密度脂蛋白胆固醇,IL-6、TNF-α、IL-1β、MDA 水平降低, p-p65 蛋白表达量减少,SOD、CAT 活性升高,且p-PI3K、p-Akt 蛋白表达量增加,差异均有统计学意义(P<0.05);PI3K/ Akt 通路抑制剂LY294002 可逆转电针对高脂饮食诱导肥胖小鼠上述指标的改善作用(P<0.05)。【结论】电针足三里、天枢、 三阴交、中脘穴可改善糖脂代谢紊乱和氧化应激治疗高脂饮食诱导的小鼠肥胖,作用机制可能与调控PI3K/Akt/核转录因子 κB(NF-κB)通路有关。

[Abstract]
Objective To observe the therapeutic effect and mechanism of electroacupuncture at Zusanli(ST36), Tianshu(ST25),Sanyinjiao(SP6),and Zhongwan(CV12)on obese mice induced by a high-fat diet. Methods An obese mouse model was established by feeding a high-fat diet. The mice were randomly divided into a model group,an electroacupuncture group(acupuncture at Zusanli,Tianshu,Sanyinjiao,and Zhongwan),and an electroacupuncture + inhibitor group(acupuncture at Zusanli,Tianshu,Sanyinjiao,and Zhongwan combined with intraperitoneal injection of LY294002). Mice fed a normal diet served as the normal control group. After the intervention,body weight was measured in each group. Fasting blood glucose,total cholesterol,triglycerides, and low-density lipoprotein cholesterol were measured using an automatic biochemical analyzer. Liver tissue morphological changes were observed by hematoxylin-eosin(HE)staining. Hepatic levels of pro-inflammatory factors interleukin-6(IL-6),tumor necrosis factor-alpha(TNF-α),interleukin-1 beta(IL-1β),and lipid peroxidation product malondialdehyde(MDA)were detected by enzyme-linked immunosorbent assay(ELISA). Hepatic antioxidant enzymes superoxide dismutase(SOD)and catalase(CAT)activities were measured using commercial kits. The protein expression levels of phosphatidylinositol 3-kinase(PI3K), phosphorylated (p)-PI3K,protein kinase B(Akt),p-Akt,p65,and p-p65 in liver tissue were detected by Western Blot. Results Compared with the normal control group,the model group showed severe disruption of liver tissue structure,with intracellular vacuoles and swelling in hepatocytes. Body weight,fasting blood glucose,total cholesterol,triglycerides,low-density lipoprotein cholesterol,levels of IL-6,TNF-α,IL-1β,MDA,and p-p65 protein expression were significantly increased,while SOD and CAT activities and p-PI3K and p-Akt protein expression were significantly decreased(P<0.05). Compared with the model group, the electroacupuncture group exhibited significantly improved liver tissue structure,with reductions in body weight, fasting blood glucose,total cholesterol,triglycerides,low-density lipoprotein cholesterol,levels of IL-6, TNF- α,IL-1β,MDA,and p-p65 protein expression,along with increased SOD and CAT activities and elevated p-PI3K and p-Akt protein expression,all showing statistically significant differences(P<0.05). The PI3K/Akt pathway inhibitor reversed the ameliorative effects of electroacupuncture on the aforementioned parameters in obese mice induced by a high-fat diet(P<0.05). Conclusion Electroacupuncture at Zusanli, Tianshu,Sanyinjiao,and Zhongwan can improve glucose and lipid metabolism disorders and oxidative stress thus to mitigate obese induced by a high-fat diet in mice,and the underlying mechanism may involve the regulation of the PI3K/Akt/NF-κB pathway.
